2. A mother has reported to A&E because her 6 month old daughter (Patient A) is presenting with fever, runny nose, barky cough, noisy and harsh breathing, and a decreased appetite. The treating physician takes a nasopharyngeal swab and sends it to Clinical Microbiology for a full respiratory virus panel. Within 24 hours the following results arrive (Figures 1-3). All results are based on the AusDiagnostics Respiratory Pathogen Multiplex RT-PCR assay with a Cycle Threshold (Ct) of 0.03. 
a. The laboratory staff have come to you with the Respiratory Pathogen assay results (Figure 1) i. Laboratory staff are confident with a positive Seasonal Coronavirus PCR result but are unsure whether the Parainfluenza 1-3 result is reliable. Describe and compare the amplification and melt curves for Seasonal Coronavirus and Parainfluenza and explain why staff are not confident about Parainfluenza 1-3 result (30 marks) ii. Explain the importance of including sample adequacy and spike samPleS to a run (as seen in Figure 1) and why not including them could cause significant problems for reporting PCR results (30 marks) 
b. Figure 2 shows the Positive Control from a Respiratory Pathogen PCR run and Figure 3 shows the results of one patient on the same PCR run. Explain the importance of positive controls when performing multiplex PCR and how the results from the positive control panel (Figure 2) affect the reporting of results for Patient B.
